I’m in Draw mode with simple brush; zbrush always starts with the same default Draw Size, Focal Shift, and RGB Intensity. Is there a way to change these defaults so that when I start Zbrush again the settings are where I left them?

Try looking for a text document in the ZData or ZStartup folder. If you find one hit ctrl+f and look for the values you mentioned, and if it has something along the lines of “Intensity : 25” then you should try to change it. (Make a backup of that text file before you do, though)
The only files in those directories with an “intensity” in them are binary so can not be edited.

I haven’t messed with this plug-in yet, but it might be handy:

And maybe there are startup scripts or macros that will auto do stuff for you. Either way, you might always be required to click a button after loading zbrush.
Never, ever, EVER edit the contents of the ZData folder. Doing so can destabilize ZBrush.
Another thing you can do would be to record a macro where you set the values that you want. You can even assign a hotkey to that macro so that you can launch ZBrush, then press a single key to get it set up the way you want it.
